A Study on the Development of Norepinephrine Supersensitivity by Conaine



Abstract
The influence of cocaine on the response of isolated rat vas deferens to norepinephrine, oxymetazoline and angiotensin was examined.
The results were as follows
1 . Cocain produced a significant elevation of maximum response to norepinephrine as well as left shift of the dose-response curve.
2 Cocaine produced a shift to the dose-reponse curve and an increase of maximum response to oxymetazol ine.
3 . Potentiation of norepinephrine effect induced by cocaine was inhibited by verapamil.
4. Cocaine had no effect on the dose response curve to angiotensin.
The results lead to the conclusion that the posssible existence of at "least two different mechanisms for the potentiation of norepinephrine effect on the isolated rat vas deferens. One is presynaptic inhibition of norepinephrine uptake, and the other is postsynaptic mechanism probably related to the improvement of intracellular calcium availability.
